A comparison of the DNA from the three collections
(RET 629-2, RET 637-3, Mycomap 9697)
was made by alignment of the sequences.
In addition to the
sequence segment posted by Stephen, Karen Hughes and
Linas Kudzma (Univ. Tenn., Knoxville) also produced
relevant sequences. If we look at all the data
we have data running from within the ITS1F primer
site in nrSSU into nrLSU. The overall aligned
region is 746 characters long.
The 5' terminus of nrITS falls at character position
76. (There are gaps, etc. caused by untrimmed
ends of data files, etc.).
The 3' terminus of nrITS falls at character
Stephen's posted sequence runs between these
(beginning/end) character positions: 29 and 661 and
includes the entire nrITS for his material.
A rough characterization of the sequences based on
the alignment follows:
char. 143 is a G in the previous RET 629-2 data, but
an A in the two sets of data for the Ontario material.
At around char. 250 there is a multi-"A"
repeat. The length differs in RET 629-2
relative to the other data by a single
character. This could occur naturally or in
the process of copying the DNA for sequencing.
It is a difference without any weight to it.
char. 277 is a C while in the other data there is a
clear T (although sometimes with a T combined with a
C as a distinctly lesser peak. Maybe a SNP?
The same is true for char. 288.
Char. 304 is an A in the sequence in
question.&nbssp; It is a G in all the other data
except in the 3' portion of some data that is beginning to deteriorate in terms of signal quality.
That is the complete list of differences.
Summary: At minimum 2 hard differences and
two possible differences (if the SNP suggestions
don't hold up to further examination.
The Thornhill material (RET 637-3) was briefly treated

Each spore data set is intended to comprise a set of measurements from a single specimen made by a single observer;
and explanations prepared for this site talk about specimen-observer pairs associated with each data set.
Combining more data into a single data set is non-optimal because it obscures observer differences
(which may be valuable for instructional purposes, for example) and may obscure instances in which
a single collection inadvertently contains a mixture of taxa.
